Appearance
This product is a white to light yellow or greenish yellow freeze drying solids.
Pharmacologic action
Pemetrexed through transport carrier of folic acid and folic acid binding protein on the
cell membrane transport system into the cell. Once the pemetrexed enters the cells, it is in
the leaves under the action of acyl glutamate synthetase more changes into the form of
glutamic acid. Glutamate persist more cells become thymidylic acid synthetase and ammonia
amide nucleotide formyl transferase inhibitor. More pancreatic acidification in rendering
time – concentration dependence process within the tumor cells, and in normal tissues
concentration is very low. More pancreatic acidic metabolites in the half-life of extended
within the tumor cells, and thus prolong the effect of drug in tumor cells. Preclinical studies
have shown that pemetrexed inhibits tumor cell lines in vitro (MSTO? 211 h, NCI? H2052)
growth. Mesothelioma cell line MSTO? 211 h research shows pemetrexed have synergy with cisplatin plus.
Genetic toxicity
In mice bone marrow micronucleus measurement display pemetrexed is breaking agent, but several
in vitro experimental study (Ames determination, determination of CHO cells) are not shown mutagenic effect.
Reproduction toxicity
Pemetrexed according to 0.1 mg/kg/day or larger doses (equivalent to 1/1666 of the recommended dose)
human male mice, too little can lead to reduced fertility, semen and testicular atrophy.
Carcinogenesis
